Isolation and identification of a potent lytic phage from Escherichia coli fermentation

Abstract: There are many kinds of phage in nature and industrial fermentations are very susceptible to phage infection.In this study,a lytic phage was isolated from culture broth of industrial threonine producing bacterium(derivative of Escherichia coli MG1655).The phage were isolated and purified by double-layer agar plate method,and plaque characteristics of the phage were observed.Morphological analysis was performed by electron microscope.A new lytic phage of Escherichia coli MG1655 was isolated.The phage plaque was big,circular,transparent and neat. Morphological analysis by electron microscopy revealed that the phage had an icosahedron head and a long tail.The results of restriction enzyme digestion indicated that the nucleic acid type of the phage was dsDNA,Based on these results,the phage was identified as a member of the long-tailed phage family named as MHV4.These results laid the foundation for the screening and transformation of the subsequent anti-phage strain for threonine production.
