Abstract: 为了建立L—谷氨酰胺发酵生产的实用方法,采用我们以前报道的一株产L—谷氨酰胺突变株里加黄杆菌708(Flavobacterium rigense)研究了大量积累L—谷氨酰胺的培养条件。结果在含有葡萄糖、酵母膏、延胡索酸铵、KH2PO4、K2HPO4·MgSO4·7H2O和CaCO3(PH6.4)的培养基中培养48小时后,谷氨酰胺的产量为25毫克/毫升。L—谷氨酰胺的积累取决于延胡索酸铵的浓度。在较高延胡索酸铵浓度时的亚适生长对高产L—谷氨酰胺是必需的。在最适条件时,谷氨酸作为一种副产物形成,其浓度少于1毫克/毫升。但其它氨基酸的积累是很少的。从培养液中分离得到的产物容易用阴离子交换层折纯化。得到产量80%的L—谷氨酰胺纯结晶均为旋光纯和层折纯。
