Abstract: 目的 观察胰岛素对小鼠早期胚胎发育以及胚胎组织中IGF-1表达的影响.方法 将ICR小鼠2-细胞胚胎在体外通过KSOM培养液培养36h,并在培养液中添加O.25μg/ml胰岛素.将添加和没有添加胰岛素体外培养到囊胚的胚胎分别转入受体母鼠继续发育,等F1代小鼠出生后比较两组间的体重差异,并对出生F1代小鼠三种组织(肝、肌、脑)中胰岛素样生长因子-1(IGF-1)的mRNA表达量进行半定量RT-PCR检测.结果 添加胰岛素培养的小鼠胚胎其囊胚发育率平均比对照组高9.7%(P<0.01).而且实验组F1代小鼠出生体重平均较对照组高17.8%(P<0.01).半定量检测发现F1代的出生小鼠肌肉中,IGF-1表达水平高于对照组.而肝脏和脑中表达没有差异.结论 适量的胰岛素环境具有加快胚胎发育和刺激出生小鼠肌肉中IGF-1表达的作用.
